"Adherent-invasive E. coli, or AIEC, are bacteria that are abnormally abundant in the gut of individuals with Crohn’s disease, a chronic inflammatory disease that affects the lining of the gastrointestinal tract. To determine whether targeting AIEC could help relieve symptoms of Crohn’s disease, researchers recently explored the possibility of simply making these bacteria less sticky. The team applied a compound known as TAK-018 to gut samples gathered from patients. TAK-018 binds to the bacterial adhesin FimH, blocking bacteria clinging and interaction with cells along the intestinal wall. Tests revealed that TAK-018 not only prevented AIEC bacteria from adhering to intestinal tissue but also reduced inflammation, helping preserve the integrity of this important barrier. While more work is needed to understand how TAK-018 operates in the body, the drug is known to be safe and well-tolerated in patients and is currently undergoing phase II testing for the treatment of Crohn’s disease..."

"Exosomes are small membrane-bound vesicles that facilitate cell-to-cell communication by transporting biomolecules like proteins, RNA, and lipids. Due to their ubiquity and cargo-carrying abilities, exosomes have many potential uses in clinical medicine. First, they can be found in many biofluids like blood, urine, cerebrospinal fluid, saliva, and milk. This means they’re easy to collect and could be used in diagnostic testing as biomarkers. Beyond that, exosomes can be used to deliver cargo to key cells, either using naturally occurring exosomes or fashioning them into drug delivery vehicles. There are even promising results for exosome-based vaccines, not only for infectious diseases but anti-tumor vaccines as well. While the field has developed rapidly in recent decades, there is much more work to do. Specifically, researchers still need to elucidate the mechanisms of exosome transfer within the body..."

EVs carry various biomolecules including proteins, DNA, RNA, and lipids and play critical roles in intercellular communication, including influencing the behavior and function of recipient cells. EVs have great potential in the clinical environment as diagnostic markers, treatment delivery vehicles, or therapeutic targets. However, to best utilize them researchers need to understand the mechanisms influencing EVs. Significant progress has been made in understanding the factors that regulate communication between cells via EVs, but there is still much to learn about what regulates EV targeting and uptake by recipient cells. Pancreatic cancer is a devastating disease with an estimated 5-year survival rate of only 11%. Most cases, over 80%, are not found until the cancer is too advanced to successfully treat, but pancreatic cancer patients show shifts in their oral microbiome, which could be detected years earlier than current methods allow.
