Students build small-sized prototypes of mountain rescue litters rescue baskets for use in hard-to-get-to places, such as mountainous terrain to evacuate an injured person (modeled by a potato) from the backcountry. Groups design their litters within constraints: they must be stable, lightweight, low-cost, portable and quick to assemble. Students demonstrate their designs in a timed test during which they assemble the litter and transport the rescued person (potato) over a set distance.

Topics include planar and spatial kinematics, and motion planning; mechanism design for manipulators and mobile robots, multi-rigid-body dynamics, 3D graphic simulation; control design, actuators, and sensors; wireless networking, task modeling, human-machine interface, and embedded software. Weekly laboratories provide experience with servo drives, real-time control, and embedded software. Students will design and fabricate working robotic systems in a group-based term project.

"RNA base editing has great potential for use in research for cell functions and genetic disease. Unlike DNA editing, which can cause unwanted mutations in other parts of the genome, RNA editing may allow researchers to mimic genetic variants that provide a health advantage. Two main RNA base editors have been used in vitro: REPAIR, which mediates A-to-I editing, and RESCUE, which can perform both C-to-U and A-to-I editing. Unfortunately, although RESCUE is more versatile, its low editing efficiency limits its applications. Now, researchers have developed an enhanced RESCUE (eRESCUE) system. eRESCUE was generated by fusing inactivated PspCas13b with ADAR2. In tests using human cell lines, eRESCUE mediated more efficient A-to -I and C-to-U RNA editing than the original RESCUE editor. eRESCUE editing of IKKβ successfully converted 177Ser to Gly, resulting in decreased phosphorylation and downregulation of downstream IKKβ-related genes..."

"RNA base-editing tools enable scientists to study gene function and develop treatments for genetic diseases. The pioneering tool REPAIR changes A bases into I bases, while the newer RESCUE system changes A and C bases into I and U bases, respectively. However, both systems tend to edit unintended, or off-target, sites in addition to desired sites. To enable more precise RNA editing, researchers recently designed a new system, ecRESCUE, by fusing the destabilization domain (DD) of the E. coli protein ecDHFR to the RESCUE system. The DD degrades the two RESCUE proteins (dRanCas13b and ADAR2) unless the drug TMP is present, making the system drug-inducible. In vitro, ecRESCUE was just as effective as RESCUE in the presence of TMP, but ecRESCUE-mediated editing was greatly reduced in the absence of TMP. When 3 specific sites were targeted, TMP-induced ecRESCUE made many fewer off-target edits than RESCUE..."
